Function

Sodium Potassium ATPase alpha 1 (ATP1A1) belongs to the P-type ATPase family and is the catalytic component of the active enzyme, which catalyzes the hydrolysis of ATP coupled with exchange of sodium (Na+) and potassium (K+ ) ions across the cellular plasma membranes. This action creates an electrochemical gradient of Na+ and K+ ions, providing the energy for active transport of various nutrients. Its phosphorylation at Tyr-10 position alters the pumping activity whereas dephosphorylation mediated by PP2A (protein phosphatase 2A) following increases in intracellular Na+, leads to catalytic activity. Because the plasma membrane is highly permeable to water, it is the concentration of ions across this membrane that is critical for maintaining an adequate cell volume and sodium potassium ATPase is key player in this maintainance process because it provides the driving force for active Na+ as well as K+ transport into/out of the cell, with water following isosmotically. This pump is consists of equimolar ratios of two main subunits, the catalytic alpha subunit (which contains the binding sites for the mentioned cations, ATP, and cardiotonic steroid inhibitors) and the regulatory beta polypeptides (which is essential for the normal activity of the enzyme as well as involved potentially in the occlusion of K+ and modulation of the enzyme's K+ /Na+ affinity).
